The receptor tyrosine kinase gene linotte is required for neuronal pathway selection in the Drosophila mushroom bodies

Abstract: The linotte (lio) mutant was first isolated as a memory mutant. The lio gene encodes a putative receptor tyrosine kinase (RTK), homologous to the human protein RYK. This gene has been independently identified in a screen for embryonic nervous system axonal guidance defects and called derailed (drl). Here, we report that linotte mutants present structural brain defects in the adult central complex (CX) and mushroom bodies (MB). linotte and derailed are allelic for this phenotype, which can be rescued by a drl+ … Show more

“…We screened 30 GAL4 drivers for expression in larval MB KCs (O'Dell et al, 1995;Yang et al, 1995;Connolly et al, 1996;Koushika et al, 1996;Amrein and Axel, 1997;Tettamanti et al, 1997;Kraft et al, 1998;Moreau-Fauvarque et al, 1998;McBride et al, 1999;Hayashi et al, 2002;Manoli et al, 2005;Keleman et al, 2007;Krashes et al, 2007;Tanaka et al, 2008). In addition, we screened another 64 GAL4 drivers (Tanaka et al, 2008) for expression in non-KC MBINs or in MBENs.…”

“…Although several mutants with MB midline crossing defects have been described (Boquet et al, 2000b;Moreau-Fauvarque et al, 1998;Simon et al, 1998), a detailed time-course that could distinguish pathfinding defects from later onset defects has been reported only for fmr1 mutants, defective in the Drosophila homolog of the fragile X mental retardation gene (Michel et al, 2004). In fmr1 mutants, ␣/␤ axons extend branches across the midline by 24 hours PPF and medial lobe fusion appears complete by 48 hours PPF, consistent with a defect in initial outgrowth (Michel et al, 2004).…”

“…First, prevention of mushroom body development results in poor courtship conditioning memory [McBride et al, 1999]. Second, the Drosophila memory mutant linotte has the same mushroom body phenotype [Moreau-Fauvarque et al, 1998]. Third, the ␣/␤ lobes are required for memory retrieval [McGuire et al, 2001].…”
