The receptor TREML4 amplifies TLR7-mediated signaling during antiviral responses and autoimmunity

Abstract: The genes and pathways that fine-tune TLR7-mediated innate inflammatory responses remain to be fully elucidated. Using an unbiased genome-scale shRNA screen, we identified the receptor TREML4 as an essential positive regulator of TLR7 signaling. Macrophages from Treml4–/– mice were hyporesponsive to TLR7 agonists and failed to produce type I interferon due to impaired phosphorylation of the transcription factor STAT1 by the MAP kinase p38 and decreased recruitment of MyD88 to TLR7. TREML4 deficiency reduced pr… Show more

“…TREML4 is expressed on CD24 + cDCs, monocytes (Hemmi et al, 2012) and macrophages, where it regulates TLR7 signaling amplification (Ramirez-Ortiz et al, 2015). Ly-6C hi monocytes were heterogeneous for TREML4 expression, but Ly-6C lo monocytes were uniformly TREML4 positive (Fig.…”

“…Multiple PRRs work in synergy to detect viral RNA or proteins synthesized during influenza replication (4648). A well-orchestrated host response may help to maintain the inflammatory response while inducing an effective adaptive immune response for viral clearance.…”

“…At day 9, BMDCs were stimulated with 100 ng/ml LPS and after 30min Brefeldin A (GolgiPlug, BD Biosciences) was added. After 8h of LPS stimulation, cells were harvested, fixed and stained for Tnf (Ramirez-Ortiz et al, 2015) and Cd11c, and FACS sorted (Supplemental Experimental Procedures). The genome-wide screens were performed as three independent replicates; in the first screen, 60 million infected cells yielded 350 million cells at day 9, while in the second and third screens, 200 million infected cells yielded 1 billion BMDCs at day 9.…”
