The purification, identification and bioactivity study of a novel calcium-binding peptide from casein hydrolysate

Abstract: In this study, a novel calcium-binding peptide from casein hydrolysate was purified using reversed-phase high performance liquid chromatography and sequenced by high-performance liquid chromatography-mass spectrometry (MS)/MS.

“…Zhang identified the novel calcium-binding peptide KGDPGLSPGK from Pacific cod bone gelatin hydrolysate and verified that Asp-3 and Lys-10 are calcium-binding sites. A novel calcium-binding peptide, VLPVPQK, identified from casein, effectively promotes the transport and absorption of calcium ions, and also contains Val, Leu, Pro, and Lys [ 55 ]. Calcium-binding peptides extracted from bovine serum have also been confirmed to contain Lys.…”

“…For example, EPQWFL derived from microalgae, VLPVPQK derived from milk, and RVYFFKGKQYWE derived from vitronectin have been shown to have superior osteogenic activity. 35–39 In addition, peptides containing glutamine sequences have been identified in some endogenous osteogenic proteins, such as GQGFSYPYKAVFSTQ derived from BMP and ALKRQGRTLYGFGG from OPG. 40 In this study, the content of glutamic acid/glutamine in SCIP was 21.1%, which is close to the recognized high-quality proteins of wheat and corn.…”

“…Calcium chelating activity was determined by the o-cresol phthalein colorimetry method ( 15 ). Five milligrams of CPPs freeze-dried powder were mixed with 1 mL of 5 mM CaCl 2 solution and 2 mL of phosphate buffer.…”

“…For example, Liu et al found that CPPs can promote the transport of calcium in Caco-2 monolayers ( 14 ). Liao et al purified and identified a novel calcium-binding peptide with good calcium transport capacity from casein hydrolysate ( 15 ). Studies have shown that CPPs have good osteogenic activity.…”
