“…In addition, fyn participates in the cell cycle (Yasunaga et al, 1996;Sette et al, 2002). Together with the data of Shirazi and Wood (1993) demonstrating the upregulation in fyn in AD brain, our present studies showing the presence of a fyn phosphorylated site in PHF tau further support a role for fyn in neuropathogenesis.…”

“…Previously, we reported that tau is tyrosine phosphorylated when cotransfected with fyn (Lee et al, 1998), a src family tyrosine kinase that has a role during the development of the nervous system (for review, see Maness, 1992;Thomas and Brugge, 1997). The possible relevance of fyn to neurodegeneration has been suggested by Wood and coworkers, reporting that neuritic plaques and dystrophic neurites in AD brain contained phosphotyrosine (Wood and Zinsmeister, 1991) and that fyn was upregulated in a subset of neurons that also stained for abnormally phosphorylated tau (Shirazi and Wood, 1993). In addition, several other studies have suggested a role for tyrosine phosphorylation in AD.…”

“…Fyn immunoreactivity (IR) is increased in AD brains (Shirazi and Wood, 1993), and genetic ablation of Fyn rendered hippocampal slices resistant to the neurotoxic effects of A␤ oligomers (Lambert et al, 1998). Here, we used genetic modulations of Fyn (overexpression vs ablation) to test the hypotheses that Fyn is involved in degenerative (synaptic deficits) and trophic (aberrant sprouting) effects of hAPP/A␤.…”

“…Indeed, Fyn activation occurs during engagement of integrins and glutamate receptors (Roskoski, 2004;Salter and Kalia, 2004), both of which have been implicated in A␤-induced disruption of synaptic function (Klein et al, 2001;Grace et al, 2002;Williamson et al, 2002). Furthermore, the distribution and levels of Fyn are altered in AD brains (Shirazi and Wood, 1993;Ho et al, 2004), and the toxic effects of A␤-derived diffusible ligands on murine hippocampal slices were blocked by the genetic ablation of Fyn (Lambert et al, 1998). Finally, ablation of Fyn decreased, whereas overexpression of Fyn increased, A␤-induced synaptotoxicity and premature mortality in human APP (hAPP) mice (Chin et al, 2004).…”
