The protein pattern of PMF virus, a type-D retrovirus from malignant permanent human cell lines

Abstract: The protein pattern of a type-D retrovirus (PMFV) isolated from and propagated in human cell lines has been investigated using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Staining with Coomassie blue and labeling with 14C-leucine/14C-lysine revealed five viral polypeptides with molecular weights of 10,000, 12,000, 15,000, 25,000, and 68,000. The 68,000 D-protein was shown to be a glycoprotein by incorporation of 3H-glucosamine and the 15,000 D-protein was identified as a phosphoprotei… Show more

“…1 shows the separation of proteins of purified t y p e D retroviruses after electrophoresis in a 7.5 to 15 per cent SDS polyacrylamide gradient slab gel. The patterns are similar to previously published d a t a for these viruses (4,18,19,24,27). A l t h o u g h the smaller proteins of each virus are designated as p l0, p12 and p15, respectively, it can be seen t h a t the molecular weights of these proteins differ slightly.…”

“…They were labelled with 14C amino acid mixture (2 t~Ci/ml, The Radiochemical Centre Amersham, specific radioactivity 58 Ci/atom carbon) and ~H glucosamine (10 txCi/ml, The Radiochemical Centre Amersham, specific radioactivity 2.8 Ci/mmol) as described (25). Radiolabelling with 3:p phosphoric acid was performed according to S~ et al (21).…”

“…As with SFV and SIV, rare cases of genuine infection of primate handlers with SRVs have been described (282). In addition, SRVs grow efficiently in many human cell lines, and a number of reports that claimed the isolation of new human retrovirus turned out to be cases of laboratory contamination with SRV (19,262,496).…”
