The (pro)renin receptor and body fluid homeostasis

Abstract: The renin-angiotensin system (RAS) has long been established as one of the major mechanisms of hypertension through the increased levels of angiotensin (ANG) II and its resulting effect on the sympathetic nerve activity, arterial vasoconstriction, water reabsorption, and retention, etc. In the central nervous system, RAS activation affects body fluid homeostasis through increases in sympathetic nerve activity, water intake, food intake, and arginine vasopressin secretion. Previous studies, however, have shown … Show more

“…PRR is shown to be colocalized to the paraventricular and supraoptic nuclei of human hypothalamic with AVP 42 and functionally regulate central AVP production. 17,43,44 As expected, AVP level was unaffected by intrarenal PRR inhibition or CD PRR deficiency.…”

“…Pro‐renin, a critical chaperone for the assembly of H + ‐ATPase subunits, regulates cell proliferation via the mitogen activated protein kinases ERK1/2 cascade and is a crucial component of Wnt pathways (Kaneshiro et al. ; Cao and Feng ; Seva Pessoa et al. ; Sihn et al.…”

“…We were intrigued by the H + -ATPase in the intercalated cells because in the kidney, the prorenin receptor (PRR) co-localizes with H + -ATPase in intercalated cells and functions as a receptor for renin. Pro-renin, a critical chaperone for the assembly of H + -ATPase subunits, regulates cell proliferation via the mitogen activated protein kinases ERK1/2 cascade and is a crucial component of Wnt pathways (Kaneshiro et al 2006;Cao and Feng 2013;Seva Pessoa et al 2013;Sihn et al 2013;Danser 2015;Ramkumar and Kohan 2016a,b;Peters 2017). There is robust and uniform PRR expression on the apical membrane of cyst epithelium in 11 weeks old Aqp2-CreTsc2 mice (Fig.…”

“…The renin-angiotensin system (RAS), as a major mechanism contributing to the regulation of blood pressure, body fluid, and tissue remodeling, has a critical role in the maintenance of tissue perfusion and homeostasis [1–3]. Seminal studies demonstrating the existence of an inhibitory arm of the RAS through the expression of angiotensin-(1-7) and angiotensin converting enzyme 2 (ACE2) [4] are now expanded with the discovery of an extended form of angiotensin I (Ang I) [5] which our laboratory characterized as an intracrine tissue precursor for direct angiotensin II (Ang II) production [68].…”
