The Primary Role in Biologic Activity of the Neutrophil Chemokines IL-8 and GRO-αin Cultured Nasal Epithelial Cells

Abstract: Primary nasal epithelial cells were investigated for their ability to synthesize and deliver neutrophil chemotactic factors (chemokines) following tumor necrosis factor-alpha (TNF-alpha) induction. The chemokines interleukin8 (IL-8), growth-related oncogene-alpha (GRO-alpha), epithelial cell-derived neutrophil attractant-78 (ENA-78), and granulocyte chemotactic protein-2 (GCP-2) have been detected and characterized and shown to have different potencies in the chemotaxis of neutrophils. Cultures of primary nasa… Show more

“…The medium was exchanged by serum-free medium 24 h prior to stimulation. The epithelial phenotype of cells was confirmed by staining of epithelial cells with a monoclonal anti-pan cytokeratin antibody as previously shown [13]. A549 cells (ATTC CCL-185) were obtained from the American Type Culture Collection (Manassas, Va., USA) and cultivated as described previously [14].…”

“…The tissue was washed with PBS and incubated with trypsin (0.5%) overnight at 4°C, as previously described, with minor modifications (13). Cells were grown to confluence (ϳ2 wk) and passaged two further times before they were used for experiments.…”

“…In addition to GROs, this family also comprises epithelium-derived neutrophilactivating peptide 78 (CXCL5), granulocyte chemotactic protein 2 (CXCL6), neutrophil-activating peptide 2 (CXCL7), and IL-8 (CXCL8) (8,9). CXCL1, CXCL2, and CXCL3 are well known as powerful neutrophil chemoattractants (10) and are involved in cancer metastasis (11), angiogenesis, and wound healing (12). Recently, GROs have been postulated to play a role in asthma severity (13,14) and asthmatic airway remodeling (7,15).…”
