Divergence of chemical plant defence mechanisms within the Brassicaceae can be utilized to identify means against specialized pest insects. Using a bioassay‐driven approach, we (a) screened 24 different Brassica napus cultivars, B. napus resyntheses and related brassicaceous species for natural plant resistance against feeding adults of the pollen beetle (Brassicogethes aeneus), (b) tested for gender‐specific feeding resistance, (c) analysed the flower bud metabolomes by a non‐targeted approach and (d) tested single candidate compounds for their antifeedant activity. (a) In no‐choice assays, beetles were allowed to feed on intact plants. Reduced feeding activity was mainly observed on Sinapis alba and Barbarea vulgaris but not on B. napus cultivars. (b) Males fed less and discriminated more in feeding than females. (c) Correlation of the metabolite abundances with the beetles' feeding activity revealed several glucosinolates, phenylpropanoids, flavonoids and saponins as potential antifeedants. (d) These were tested in dual‐bud‐choice assays developed for medium‐throughput compound screening. Application of standard compounds on single oilseed rape flower buds revealed highly deterrent effects of glucobarbarin, oleanolic acid and hederagenin. These results help to understand chemical plant defence in the Brassicaceae and are of key importance for further breeding strategies for insect‐resistant oilseed rape cultivars.

The cabbage stem flea beetle (CSFB), Psylliodes chrysocephala L. (Coleoptera: Chrysomelidae), is one of the most important pests in European winter oilseed rape production. Adult beetles feed on young leaves whereas larvae mine within the petioles and stems. Larval infestation can cause significant crop damage. In this study, the host quality for CSFB of four oilseed rape (Brassica napus L.) cultivars and seven other brassicaceous species with different glucosinolate (GSL) profiles was assessed under controlled conditions. Larval instar weights and mortality were measured after 14 and 21 days of feeding in the petioles of test plants. To study the impact of GSL on the performance of larvae, the GSL contents in petioles from non-infested and infested plants were analysed before, and 21 days after, the start of larval infestation. Larval performance was not significantly different between the four cultivars of oilseed rape, but differed considerably among the other brassicaceous species tested. In comparison to the weight of larvae in the standard B. napus cv. Robust, the larval weight was higher in turnip rape (Brassica rapa L. var. silvestris) and significantly reduced in white mustard (Sinapis alba L.), oil radish (Raphanus sativa L. var. oleiformis), and cabbage (Brassica oleracea L. convar. capitata var. alba). The duration of larval development increased in white mustard and oilseed radish. The GSL profiles of the petioles showed little difference between non-infested and infested plants of oilseed rape whereas the content of aliphatic GSL increased in the infested turnip rape plants. In contrast, the aliphatic and benzenic GSL decreased in infested Indian rape (B. rapa subsp. dichotoma Roxb.). Larval weight was not correlated with the total GSL content of plants, neither before infestation nor 21 days after. Larval weight was positively correlated with progoitrin and 4-hydroxyglucobrassicin. White mustard, which provides inferior host quality for larval development, has the potential to introduce insect resistance into high-yielding oilseed rape cultivars in breeding programmes.

Dealing with insect pests is one of the biggest challenges facing the cultivation of oilseed rape (Brassica napus, OSR). Insect pests are usually controlled using insecticides, but the increasing occurrence of insecticide-resistant populations and the socio-economic context argue against the sole use of these substances. Plant resistance is a classical and proven alternative crop protection strategy that is the basic tool of integrated pest management. However, no insect-resistant OSR cultivar is currently available on the market. Here, I review some of the constraints that make phenotyping for insect resistance particularly challenging with OSR and give perspectives to develop this resistance. Some studies have proved that breeding OSR for insect resistance could be achieved, and three strategies have been considered: introducing resistance transgenes into the OSR genome, exploiting natural variation in resistance already present in B. napus and introgressing resistance from other brassicaceous species. The white mustard, Sinapis alba, seems a particularly promising source of resistance against most of the OSR insect pests.
