The Positive Effects of Different Platelet-Rich Plasma Methods on Human Muscle, Bone, and Tendon Cells

Abstract: The utilization of different PRP separations may result in a potentially beneficial effect on the clinically relevant target cells in vitro, but it is unclear which platelet concentration or PRP preparation may be optimal for the treatment of various cell types.

“…Platelet-rich plasma is another healing promotive agent that initiates cell proliferation and tissue maturation 20,23,32 . These beneficial effects have been suggested to be due to the growth factors that are delivered from the platelets 62,64 . It seems that by equipping the tissueengineered scaffolds with cells and healing promotive agents, the tissueengineered graft could be optimized to open new insights in the field of tendon reconstructive surgery and regenerative medicine 12 .…”

“…39 PRP is safe, but effectiveness remains to be unquestionably proven. Clinical studies have shown variable results, 2,16,18,28,30 and basic science and animal studies range from positive outcomes 11,20,25,35 to suggesting little benefit for some types of injury. For example, a recent article by Delos et al 8 showed no benefit of using PRP in an animal model of contusion injury.…”

“…This suggests that PRP products have a positive effect on the cells' mitogenic activity. Also, improvement in collagen production and optimization of the important collagen I/III ratio have been observed 29,31,33 . However, these positive effects and their consequences for "real life" tissue healing have yet to be identified in clinical studies 3 .…”
