The Plasma Membrane Proton Pump PMA-1 Is Incorporated into Distal Parts of the Hyphae Independently of the Spitzenkörper in Neurospora crassa

Abstract: bMost models for fungal growth have proposed a directional traffic of secretory vesicles to the hyphal apex, where they temporarily aggregate at the Spitzenkörper before they fuse with the plasma membrane (PM). The PM H ؉ -translocating ATPase (PMA-1) is delivered via the classical secretory pathway (endoplasmic reticulum [ER] to Golgi) to the cell surface, where it pumps H ؉ out of the cell, generating a large electrochemical gradient that supplies energy to H ؉ -coupled nutrient uptake systems. To characteri… Show more

“…The PVCs either lack or have small amounts of other vacuolar proteins such as carboxypeptidase Y and alkaline phosphatase. In previous reports, vacuoles have been shown to contain significant amounts of proteins that function in the plasma membrane, such as the plasma membrane proton pump (PMA-1) and two plasma membrane calcium pumps (NCA-2 and NCA-3) (13,38). These plasma membrane proteins were not observed in the PVCs.…”

“…As controls for fractionation, antibodies to the N . crassa plasma membrane ATPase ( PMA 1), which is enriched in the plasma membrane fractions, but also localizes to tubular vacuoles (Fajardo‐Somera et al ., ) and antibodies to ERV 25, which is involved in ER ‐Golgi trafficking ( T . S tarr, unpublished results) were used.…”

“…Correct integration in the selected NRAF strains was verified by PCR as described previously (Fajardo-Somera et al, 2013). Mycelium for genomic DNA extraction was grown at 28°C for 7 days on liquid VMM without shaking or light, filtered, submerged in liquid nitrogen and lyophilized.…”
