The PI3K/Akt1 pathway enhances steady-state levels of FANCL

Abstract: The Fanconi anemia pathway supports hematopoietic stem cell survival in response to inflammatory and metabolic stress. We show that polyubiquitination and proteasome degradation of FANCL is inhibited by Akt1 activation, revealing a potentially important mechanism for the maintenance of stem cell function.

“…The expression plasmids of GFP-FANCL harboring the C312,315A or C364,367A mutations were obtained from the plasmid for expressing the wild-type GFP-FANCL provided by Dr. Grover C. Bagby from Oregon Health and Science University 17 by using a GeneArt Site-directed Mutagenesis Kit (Thermo Fisher Scientific, Waltham, MA), following the vendor’s recommended procedures.…”

“…We used CacoBBe cells with stable knockdown of occludin (shOcln) generated by Buschmann and colleagues. 15 These occludin knockdown cells were recently reported to provide (i) a >90% reduction in global occludin protein expression, (ii) a reduced TER, and (iii) to have an activated leak pathway, i.e. increased passage of dextrans in the range of 3-40 kDa.…”

“…However, contrary to the aforementioned studyclaudin-2 expression was found to be reduced in these cells. 15 Though Yu et al and VanItallie et al found no correlation between occludin deficiency and basal flux of macromolecular tracers in MDCK cells they reported a different response to cytokines or to cell physiology-affecting reagents that scale in line with the occludin level in these cells. 11,13 Similar findings were reported in mice showing that cytokine-induced barrier defects depend on occludin.…”
