The aim of the studies was to investigate the effect of camel grass, lavender, patchouli, peppermint and tea tree essential oils, and their mixtures on the in vitro growth of pathogenic fungi Cladobotryum dendroides and Mycogone perniciosa, occurring in the cultivation of button mushroom (Agaricus bisporus). The mycelial growth of the tested pathogens was evaluated on PDA medium. Essential oils were added in three doses: 0.25; 0.5 and 1 mg·cm–3 of PDA medium. Camel grass and peppermint essential oils applied at the highest dose inhibited completely the in vitro growth of C. dendroides mycelium. Lavender oil used at the amount of 1 mg·cm–3 reduced the growth of the pathogen by 90 %. In the case of M. perniciosa the complete inhibition of the pathogen’s growth was observed after the addition of camel grass oil to PDA medium, irrespective of a dose, and lavender oil at the doses of 0.5 and 1 mg·cm–3. The efficacy of the tested mixtures against M. perniciosa was high. Generally, all mixtures of essential oils, irrespective of a dose, completely controlled the growth of the pathogen. The complete inhibition of the growth of C. dendroides was observed only on the medium with the addition of the mixture of camel grass and peppermint oils at the highest dose. The conducted research showed that natural essential oils due to their antifungal properties could be useful in the Integrated Disease Management for the protection of button mushroom against diseases. They could be an effective alternative to synthetic chemical fungicides.

Obtained from aromatic plants, essential oils have been utilised as natural food preservatives. Their ability to inhibit microorganism growth results from their varied chemical compositions. Furthermore, due to their classification as generally recognised as safe (GRAS) and low toxicity levels, there has been an increased study of essential oils’ compositions and antimicrobial properties against pathogens living in the human oral cavity. An initial evaluation was conducted with 11 essential oils to verify their antimicrobial activities in vitro against opportunistic pathogenic bacteria Staphylococcus aureus, Streptococcus sanguinis and Streptococcus mutans, also a model yeast Candida albicans. The antimicrobial activity was evaluated by agar disk diffusion assay, while PT kindly provided chemical composition data. Darjeeling Sembrani Aroma. The result showed that tested essential oils exhibited an inhibition zone range of 8.13 to 40.00 mm for bacteria and 7.63 to 44.04 mm for yeast. Chemical compositions of the oils revealed that the major compounds are monoterpenes, followed by sesquiterpenes and phenylpropanoids.

Abstract. Andila PS, Nugroho LH. 2022. Antibacterial and phytochemical constituent of Etlingera rubroloba A.D. Poulsen extract, an endemic ginger from Wallacea Region, Indonesia. Biodiversitas 23: 3646-3658. Etlingera rubroloba (Blume) AD Poulsen is one of the wild species of the Zingiberaceae which is a native and endemic species in the Wallacea region and has not been studied well for its biology and phytochemical potential. This study aimed to investigate the potential antibacterial activity of hexane extract from the leaves and rhizome of E. rubroloba and study their phytochemical content. The antibacterial activity test was carried out by the agar disc diffusion method against several human pathogenic bacteria and be measured for the Minimum Inhibitory Concentration (MIC) and Zone of inhibition. The phytochemical profile was tested for total phenol content (TPC), total flavonoid content (TFC) and GC-MS analysis. The study results show that TPC of E. rubroloba extracts respectively 15.68±0.80 mg Gallic Acid Equivalent (GAE)/g sample (leaves) and 11.89±0.76 mg GAE/g sample (rhizomes), while TFC value were 33.99±2.24 mg Quercetin Equivalent (QE)/g sample (leaves) and 32.46±0.67 mg QE/g sample (rhizomes). The antibacterial test revealed that E. rubroloba extracts were effective against Bacillus cereus ATCC11778 (with MIC value 0.5 mg/mL using leaves extract and 1 mg/mL in rhizomes), but it was not effective against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923 and Staphylococcus epidermidis ATCC12228. The hexane extracts of E. rubroloba contained a total of 28 and 58 compounds were identified in the leaf extract and rhizome, respectively. The dominant compounds in the leaf extract (Pentadecanoic acid, 14-methyl-, methyl ester; Methyl stearate) and in the rhizome extract (Hexadecanoic acid, ethyl ester; Isopropyl myristate; 9-Octadecenoic acid (Z)-, methyl ester; 9,12- Octadecadienoic acid, methyl ester, (E,E)-) is well known to have antibacterial activity. Based on the results of the antibacterial and phytochemical studies, it can be suggested that E. rubroloba has the potential as an antibacterial agent.
