Phylogenetic analyses of the nuclear-encoded small-subunit rDNA sequences from taxa representing all of the major lineages of green algae, including new sequences for the Trentepohliales, consistently indicated that the subaerial Trentepohliales are closely related to ulvophycean marine green algae, particularly to the siphonous and hemisiphonous orders. The presence of phragmoplast-type cytokinesis in the order Trentepohliales remains enigmatic, and it is interesting that this type of cell division is associated with terrestrial (subaerial) habits. INTRODUCTIONRecent reviews (e.g. Chapman et al., 1999;Waters & Chapman, 1996) of analyses of the nuclear-encoded smallsubunit (SSU) rDNA as well as the chloroplast-encoded large-subunit Rubisco gene (rbcL) provide support for the original suggestion, based on ultrastructural data, that there are two main lineages among the green plants (Pickett-Heaps & Marchand, 1972). One of the lineages comprises the charophycean algae and their descendents, the land plants (charophycean lineage sensu Pickett-Heaps & Marchant, 1972), forming a monophyletic group named Streptophyta (sensu Cavalier-Smith, 1993). The charophycean algae include taxa from at least five orders: the Chlorokybales, Klebsormidiales, Zygnematales, Coleochaetales and Charales. The second lineage (chlorophycean algae sensu Pickett-Heaps & Marchant, 1972) consists exclusively of the remaining green algae (containing at least Chlamydomonas and its allies, the coccoid green algae, Dasycladales, Cladophorales and Trebouxiophyceae), forming the monophyletic group Chlorophyta (sensu Cavalier-Smith, 1993;Friedl, 1997;Melkonian et al., 1995). The term 'chlorophyte' has often been used to denote all green algae; however, it should now be used only as an informal designation for green algae in the Chlorophyta [see Chapman et al. (1999) for a revisionary approach to the systematics of the green algae].Conflicting hypotheses have been offered about the systematic position of the Trentepohliales among the several classes of green algae. The presence of multilayered structures in the flagellar apparatus and the demonstration of a phragmoplasttype cytokinesis in Cephaleuros parasiticus (Chapman & Henk, 1986) and Trentepohlia odorata (Chapman et al., 2001;Waters et al., 1998) suggest an affinity with the class Charophyceae. However, taxonomic features of the Trentepohliales such as the anticlockwise flagellar apparatus components can be cited as evidence for an affinity with the Ulvophyceae (Roberts, 1984). Based on biochemical, biophysical and physiological features, Raven (1987) classified the Trentepohliales among a third class, the Pleurastrophyceae. Moreover, it was noted that the Trentepohliales even share a rare ultrastructural feature (presumptive mating structures in the gametes) with members of a fourth class, the Chlorophyceae (Chapman & Henk, 1983, 1985. Pressurized liquid extraction (PLE), an environmentally friendly technique, has been used to obtain antiviral compounds from microalgae commonly used as carotenoids sources: Haematococcus pluvialis and Dunaliella salina. The antiviral properties of PLE extracts (hexane, ethanol and water) were evaluated against herpes simplex virus type 1 (HSV-1) at different stages during viral infection. Pre-treatment of Vero cells with 75 g mL -1 of H. pluvialis ethanol extract inhibited virus infection by approx. 85%, whereas the same concentration of water and hexane extracts reduced the virus infectivity 75% and 50% respectively. D. salina extracts were less effective than H. pluvialis extracts and presented a different behaviour, since water and ethanol extracts produced a similar virus inhibition (65%). Moreover, H. pluvialis ethanol extract was also the most effective against HSV-1 intracellular replication.The antiviral activity of water PLE extracts was found to correlate with polysaccharides, since the polysaccharide-rich fraction isolated from these extracts showed higher antiviral activity than the original water extracts. A GC-MS characterization of the H.pluvialis ethanol extract showed the antiviral activity of this extract could be partially related with the presence of short chain fatty acids, although other compounds could be involved in this activity; meanwhile, in the case of D. salina ethanol extract other compounds seemed to be implied, such as: β-ionone, neophytadiene, phytol, palmitic acid and α-linolenic acid.

Recent phylogenetic studies of Chlorella-like algae revealed that this group has diversified into free-living and endosymbiotic niches, the latter within protists and invertebrate organisms. Our previous studies using a long-term culture composed of an alga (Chlorella vulgaris, rDNA sequence unknown), a bacterium (Escherichia coli) and a ciliate (Tetrahymena thermophila), suggested that some clones in the algal population developed an endosymbiotic ability with T. thermophila while others developed an ectosymbiotic ability with E. coli from a non-associated stage. In this paper, an rDNA (18S, ITS1, 5.8S, ITS2)-based phylogeny of the ancestral strain and derived clones isolated from 5-year microcosm cultures was constructed and revealed that the alga belongs to the genus Micractinium. This result was supported by analysis of the secondary structure of the rRNA gene. No difference was observed in the sequence between the ancestral and derived clones. On the other hand, some morphological and physiological traits of the ancestral strain and the derived clones were analysed and revealed that several phenotypic changes had occurred among the algal clones. In particular, the derived clones obtained from long-term cultures had increased in cell size, changed in their ability to grow at pH 4.0, and developed a tendency for cell-aggregation or sedimentation.
