The phenotypic and functional characteristics of umbilical cord blood and peripheral blood natural killer cells

Abstract: SummaryAllogeneic hematopoietic cell transplantation can be curative for patients with high-risk acute leukaemia. Umbilical cord blood (UCB) is an increasingly used source of allogeneic stem cells for patients who are in need of a transplant, but do not have a sibling donor. This review highlights the similarities and differences between the natural killer (NK) cells obtained from adult peripheral blood (PB) and UCB.

Notably, CD56 Ϫ CD16 ϩ NK cells were poorly represented in adult controls (2% Ϯ 2%; n ϭ 60), whereas, as previously described, they were consistently detectable in CB ( Figure 2B), 29,30 but their percentage was considerably lower than in patients 1, 4, 5, and 6 (Figure 2B-C).…”

“…In the present study CB the NK (CD16 + /CD56 + ) cell ratio was similar to the MPB NK cell ratio, as was previously reported for NK cells in APB and CB [36]. The nature of CB NK cells remains controversial; despite the similar frequency of NK cells in CB and APB, CB NK cell dysfunction, as compared to APB and MPB was reported.…”

“…26 Because MuHV-4 infection in mice shares important properties with Epstein-Barr virus infection in humans, we speculate that this mode of NK-cell arming may occur in healthy people. Of note, CD56 dim NK cells with cytotoxic capacity have been identified in umbilical cord blood, 27 suggesting that multiple pathways may lead to functionally competent human NK cells. Thus, some of the differences between human and SPF laboratory mouse NK-cell function may reflect a disparity in immune activation induced by chronic virus infection, as opposed to an intrinsic difference in the NK cells themselves.…”
