The pharmacological profile of ovalbumin‐induced paw oedema in rats

Abstract: Rats are commonly used in anaphylaxis models, mainly in intestinal anaphylaxis. Hypersensitivity mechanisms are complex and they are not clearly defined. Ovalbumin (OVA) is commonly used for studies on the hypersensitivity mechanism. However, the potential pro-inflammatory mediators induced by this antigen in the model of paw oedema in immunized rats are still not completely understood. This work examines the pharmacological modulation of several mediators involved in rat hind paw immune oedema induced by OVA.… Show more

“…In fact, there is substantial evidence that histamine and serotonin play important roles in the pathogenesis of allergic disorders [38,39] . For instance, histamine and serotonin signaling through H 1 and 5-HT 2 receptors, respectively, are known to be important mediators of edema formation during the immediate phase of allergic responses [40,41] . Interestingly, our data showed that although effectively precluding the increase in vascular permeability seen in the PCA, treatment with a combination of promethazine and methysergide, which block H 1 and 5-HT 2 receptors, respectively, was unable to modify the fluid consumption in OVA-immunized rats ( fig.…”

“…In active paw anaphylaxis, the edematogenic reaction is manifested early after OVA-administration and the peak effect is maintained during 30 min-1 h which decreases thereafter. It involves IgE antibodies and also biogenic amines [24,25]. Active paw anaphylaxis model has widely been utilized for screening of putative anti-allergic drugs [26].…”
