The Peroxisome Deficient Arabidopsis Mutant sse1 Exhibits Impaired Fatty Acid Synthesis

Abstract: The Arabidopsis Shrunken Seed 1 (SSE1) gene encodes a homolog of the peroxisome biogenesis factor Pex16p, and a loss-of-function mutation in this gene alters seed storage composition. Two lines of evidence support a function for SSE1 in peroxisome biogenesis: the peroxisomal localization of a green fluorescent protein-SSE1 fusion protein and the lack of normal peroxisomes in sse1 mutant embryos. The green fluorescent protein-SSE1 colocalizes with the red fluorescent protein (RFP)-labeled peroxisomal markers RF… Show more

“…Although some reports on the metabolism of seed development have been published previously they have tended to focus on earlier stages of seed development and on specific biochemical pathways largely concentrating on sugar components Baud and Graham, 2006;Gomez et al, 2006), fatty acids and oil (Cernac and Benning, 2004;Lin et al, 2004;Vigeolas et al, 2004;Gó mez et al, 2005), or specific transcriptional programs (Hobbs et al, 2004;Braybrook et al, 2006). Broader transcriptional studies have also been previously conducted with an emphasis on the stages of development from 5 to 13 DAF (Ruuska et al, 2002).…”

“…To confirm the identity of peroxisomes, a further construct was made encoding monomeric red fluorescent protein (mRFP) with a PTS1 sequence at the C-terminal end ( Figure 3A). This PTS1 comprised the 10 C-terminal amino acids of pumpkin malate synthase that had previously been verified to transport RFP to peroxisomes in Arabidopsis (Lin et al, 2004). Plants were transformed with GFP and mRFP transgenes and then crossed and fluorescence observed in the root hairs of seedlings expressing both fluorescent proteins ( Figure 3B).…”

“…Confocal microscopy was performed to observe GFP and mRFP in root hair cells as previously described (Lin et al, 2004). For transmission electron microscopy, cotyledons from seedlings germinated for 5 d on half-strength MS medium supplemented with 1% (w/v) sucrose were processed as previously described (Germain et al, 2001) …”

“…However, seed protein content was unaffected. A mutation in the Shrunken Seed1 (SSE1) gene altered seed composition by increasing starch and reducing oil and protein content (Lin et al, 2004). Mutations in metabolic pathways are also capable of dramatic changes in seed composition.…”
