The P2X7 Receptor Primes IL-1β and the NLRP3 Inflammasome in Astrocytes Exposed to Mechanical Strain

Abstract: Inflammatory responses play a key role in many neural pathologies, with localized signaling from the non-immune cells making critical contributions. The NLRP3 inflammasome is an important component of innate immune signaling and can link neural insult to chronic inflammation. The NLRP3 inflammasome requires two stages to contribute: priming and activation. The priming stage involves upregulation of inflammasome components while the activation stage results in the assembly and activation of the inflammasome com… Show more

“…ATP release through Panx1 channels has been shown to be a strong signal for the recruitment of inflammatory cells in response to apoptosis (Chekeni et al, 2010) and activation of the NLR3P inflammasome (Albalawi et al, 2017; Lappas, 2014; Wu et al, 2017). However, the precise Panx1-mediated mechanisms controlling inflammation have never been fully elucidated.…”

“…Panx1-associated ATP release increases in Caspase 1 and Pro- IL-1β processing in human gestational tissues promoting (Lappas, 2014). In astrocytes, Panx1 mediated ATP release and signaling through the P2X7 channel, has been found promote activation of the NLRP3 inflammasome and the release of IL-1β (Albalawi et al, 2017). Our results show that TNF treatments (2.5-100 ng/mL) of HUVECs resulted in the release of approximately 10-20 nM ATP, which is similar to previous reports (Tozzi et al, 2019).…”

“…In models of acute glaucoma, retinal ischemic-reperfusion (I/R) injury caused by elevated intraocular pressure (IOP) [91][92][93][94], optic nerve clamp [95], or optic nerve crush [96] resulted in upregulation of NLRP1 and NLRP3 in the retina. The resulting retinal ganglion cell loss was ameliorated in the Nlrp3 −/− mouse [97].…”

“…Due to its macromolecular nature, inflammasome assembly has been hypothesized to be facilitated by cytoskeleton reorganization (see Fig. ) . Understanding the molecular mechanisms by which mechanotransduction modulates inflammasome activation will help in developing therapies to ameliorate the above‐mentioned inflammatory diseases.…”
