The P2X4 receptor is required for neuroprotection via ischemic preconditioning

Abstract: Ischemic preconditioning (IPC), a procedure consisting of transient ischemia and subsequent reperfusion, provides ischemic tolerance against prolonged ischemia in the brain. Although the blood flow changes mediated by IPC are primarily perceived by vascular endothelial cells, the role of these cells in ischemic tolerance has not been fully clarified. In this study, we found that the P2X4 receptor, which is abundantly expressed in vascular endothelial cells, is required for ischemic tolerance following middle a… Show more

“…Such a neuroprotective response to mechanoreceptors has previously been suggested in blood‐vessels in mice (Ozaki et al. ) and in the peripheral nervous system of fish (Kolosov & Uzdensky ). Further studies are needed to determine the role of mechanoreceptors in retinal damage.…”

“…The generation of P2X4R -/mice was described before (75). The generation of P2X4R fl/fl mice was described before (76). pmeLUC transgenic mice were generated by pronuclear injection of a linearized pmeLUC DNA construct (77) into the pronuclei of B6D2F2 zygotes.…”

“…P2X4R expressed by microglia are involved in post-ischaemic brain inflammation ( Cheng et al, 2014 ). P2X4R are required for neuroprotection via ischemic preconditioning ( Ozaki et al, 2016 ). Neuronal K ATP channels play a role in hypoxic preconditioning where they decrease neonatal hypoxic-ischaemic brain injury; it has been proposed that openers of K ATP channels may prove to be therapeutically beneficial ( Sun H.S.…”
