The Origins and Functions of Tissue-Resident Macrophages in Kidney Development

Abstract: The adult kidney hosts tissue-resident macrophages that can cause, prevent, and/or repair renal damage. Most of these macrophages derive from embryonic progenitors that colonize the kidney during its development and proliferate in situ throughout adulthood. Although the precise origins of kidney macrophages remain controversial, recent studies have revealed that embryonic macrophage progenitors initially migrate from the yolk sac, and later from the fetal liver, into the developing kidney. Once in the kidney, … Show more

“…As expected, Actb was uniformly expressed in kidney and heart while Scgb1a1 and Sftpc were undetectable (Supplementary Figure 2 A, B). In both tissues, Lyz2 was expressed by a few scattered cells, which presumably correspond to macrophages (37, 38) (Supplementary Figure 2). In a subset of kidney tubular structures, we detected Napsa (Supplementary Figure 2 A), which agrees with the previously described immunohistochemical detection of the marker in renal proximal tubule cells (39).…”

“…The vast majority of our knowledge on human DCs comes from work on human peripheral blood. However, it is becoming increasingly clear that the function of DCs and many immune cells differs depending on their anatomical location (14)(15)(16)(17)(18)(19)(20)(21), which highlights the importance of studying immune cells at the site where they exert their function.…”

“…Tissue resident macrophages also play critical roles in tissue remodeling during development; the macrophages are involved in killing and removing interdigital cells in the mouse paw (Hopkinson-Woolley et al, 1994), developing the mouse eye hyaloid vessels and pupillary membrane (Bishop et al, 2016), bridging blood vessel anastomoses in the zebrafish embryo (Fantin et al, 2010), and branching morphogenesis in the murine kidney (Munro and Hughes, 2017). On the basis of the observation that the hemogenic activity of the endocardium in the heart tube coincides with the developmental remodeling of cardiac tissue, the authors hypothesized that these events might be intrinsically linked.…”
