The Original Form of C4-Photosynthetic Phosphoenolpyruvate Carboxylase Is Retained in Pooids but Lost in Rice

Abstract: Poaceae is the most prominent monocot family that contains the primary cereal crops wheat, rice, and maize. These cereal species exhibit physiological diversity, such as different photosynthetic systems and environmental stress tolerance. Phosphoenolpyruvate carboxylase (PEPC) in Poaceae is encoded by a small multigene family and plays a central role in C4-photosynthesis and dicarboxylic acid metabolism. Here, to better understand the molecular basis of the cereal species diversity, we analyzed the PEPC gene f… Show more

“…To assess how OsPRI1 and the cis ‐elements where it binds in the SvPEPC1 promoter have been modified during the evolution of C 4 photosynthesis, we first identified PEPC1 promoter sequences from two C 4 plants with independent C 4 origins ( Z. mays and S. viridis ), a C 3 member of the PACMAD clade ( D. oligosanthes ), and rice, a member of the BEP clade. For rice, the promoter of both OsPEPC1 and OsPEPC4 were included in the analysis because OsPEPC1 has been identified as the closest in sequence to C 4 PEPCs (Yamamoto et al ., 2022) and OsPEPC4 is specifically expressed in mesophyll cells (Masumoto et al ., 2010). We also identified OsPRI1 homologues in Z. mays (GRMZM2G017586 and GRMZM2G093744), S. viridis (Sevir.6G068300) and D. oligosanthes (OEL18192; Fig.…”
