The omnistat: A flexible continuous‐culture system for prolonged experimental evolution

Abstract: Microbial evolution experiments provide a powerful tool to unravel the molecular basis of adaptive evolution but their outcomes can be difficult to interpret, unless the selective forces that are applied during the experiment are carefully controlled. In this respect, experimental evolution in continuous cultures provides advantages over commonly used sequential batch‐culture protocols because continuous cultures allow for more accurate control over the induced selective environment. However, commercial contin… Show more

“…Many parts could also be easily substituted depending on availability or cost constraints. Any water-tight, sterilisable container can be used for culture chambers if suitable lids can be manufactured, such as falcon tubes (Tonoyan et al ., 2020) or commonly available lab glassware (Ekkers et al ., 2020). The controlled temperature room can be replaced with waterbaths and portable aquarium pumps can be used instead of building infrastructure.…”

“…В хемостатах удельная скорость роста регулируется за счет регулирования скорости добавления свежей среды и выведения продуктов жизнедеятельности и части клеток. В ауксостатах поддерживается стабильная плотность популяции [49]. В ретентостатах, являющихся модификацией хемостатов, происходит отток сточных вод, а биомасса задерживается с помощью специальных фильтров.…”

“…Realization of the advantages of automated (computercontrolled) continuous culturing over manual serial dilution methods of experimental evolution [6] was enabled by the recently enhanced accessibility of 3D printing technology and programmable microcontrollers (such as Arduino and Raspberry Pi). One of the rapidly emerging custom-built continuous culturing devices [7][8][9][10], termed 'morbidostat' , is a modification of the chemostat in which, instead of nutrient supply, the culture density is controlled by selective drug pressure [11,12]. This approach enables gradual genetic adaptation of evolving bacterial populations to higher concentrations of an antibiotic and generally recapitulates the knowledge of resistance mechanisms from previous studies [11,13,14].…”

“…Among recently reported continuous culturing devices, eVOLVER [ 9 ] and omnistat [ 7 ] provide technologically sophisticated multi-purpose platforms supporting a broad range of microbial evolutionary experiments in chemostat, turbidostat and morbidostat modes. The key feature of the eVOLVER platform is the capability for highly parallel processing of multiple samples over a variety of individually controlled experimental conditions.…”
