The Occurrence of the Colistin Resistance Gene mcr-1 in the Haihe River (China)

Abstract: Antibiotic failure is occurring worldwide. In a routine surveillance study on antibiotic resistance genes (ARGs) in natural water bodies, we noted the detection of colistin-resistance gene mcr-1, previously identified in Escherichia coli and Klebsiella pneumoniae isolates from human beings and animals in several countries. The mcr-1 gene might be present in water environments, because aquatic ecosystems are recognized as reservoirs for antibiotic resistant bacteria (ARB) and ARGs. In this study, a qPCR assay w… Show more

“…A systematic screening of the gene in colistin-resistant strains was performed [150] (Figure 2). For such purposes, scientists have used the primers of the original study [6], or have designed their own primers for standard PCR [132, 151160] or RT-PCR, based on SYBR Green assays [64, 151, 161], TaqMan probe [66, 72, 152, 162], or other FAM-labelled probe [9, 71, 163, 164] or HEX-labelled probe [165] (Table 4).…”

“…To date, mcr-1 has been observed on a wide variety of plasmid types, including IncI2, IncHI2, and IncX4 17 . Intensive screening efforts for mcr-1 have found it to be highly prevalent in a number of environmental settings, including the Haihe River in China 18 , recreational water at public urban beaches in Brazil 19 , and fecal samples from otherwise healthy individuals 20 , 21 . Although both Brazil and China have now banned the use of colistin in agriculture, the evidence that mcr-1 can spread within hospital environments even in the absence of colistin use 22 as well as in the community 21 raises the possibility that the spread of mcr-1 will not be contained by these bans.…”
