The Number of CD34+Cells Mobilized into the Peripheral Blood Can Predict the Quality of Subsequent Collections

Abstract: PBPC were mobilized using a variety of chemotherapy regimens plus G-CSF in a group of 126 consecutive patients. Data are presented that show a close correlation between the number of CD34+ cells mobilized into the peripheral blood (PB) and the number of CD34+ cells subsequently collected by leukapheresis (R = 0.904). On the basis of this correlation, a regression formula was calculated that could give an estimate of the total number of CD34+ cells likely to be collected by leukapheresis from a given number of … Show more

“…[11][12][13][14][15][16] Recommendations for the level of PB CD34 þ cell count that should be used to initiate apheresis has varied from 8 to 20/ml. [16][17][18][19] If harvesting is initiated at a lower PB CD34 þ count, daily yield will be lower and more apheresis days are required to achieve an adequate harvest.…”

“…[23][24][25][26] During the time period of our study, 28 patients (unpublished data) received disease-specific chemotherapy primes with G-CSF and the median time to first harvest was 13 days (range [11][12][13][14][15][16][17][18]. Stem cell recovery is more rapid however following low-dose cyclophosphamide/G-CSF.…”

“…[26][27][28] We recently achieved a yield of 14 Mks per mPB HSPC and obtained up to 45% highploidy ( > 4N) Mks. 29 This Mk productivity has the potential to allow production of one platelet transfusion from 125 million mPB CD34 + cells, but it is clear that the yield must be increased by another order of magnitude.…”

“…The immunological analysis of CD34 1 cells is currently considered the best method for identifying hematopoietic and progenitor cells and also the most precise marker to guide timing peripheral blood stem cells (PBSC) apheresis. However, it is an expensive technique and it requires technical equipment and well-trained technicians (6).…”
