The nucleotide sequence of the transforming BglII-H fragment of adenovirus type 7 DNA

Abstract: The nucleotide sequence of the leftmost BglII-H fragment (0--4.5%) of weakly oncogenic human adenovirus serotype 7 (Ad7) has been determined (1568 base pairs). This is the shortest Ad7 DNA fragment reported to transform primary rat cells into an immortal cell line (Dijkema et al., 1979). The l-strand of BhlII-H was found to contain the complete information for a polypeptide of at most 28 051 daltons, followed by the putative promoter site of the next gene. Comparison of the determined Ad7 sequence with that of… Show more

“…Later subcloning of this mutant's cDNA in an expression vector revealed modestly diminished transactivation of the Ad3 ElA-CAT reporter plasmid. The four 12S and four 13S clones each have intron junctions which agree precisely with the junctions reported for the closely related ElA gene of adenovirus type 7 (7).…”

“…The ratio of yields of the larger and the smaller products, estimated by ethidium fluorescence in the gel, is typically about 10:1. The lengths are those expected for the Ad3 ElA cDNA products, predicted from the Ad3 DNA sequence, the specific oligomer primers, and the intron junctions described for Ad7 ElA mRNA (7). Treatment with BamHI and HindlIl reduces the lengths of the two Ad3specific products by about 75 bp each.…”

“…1) requires isolation of an appropriate Ad7a DNA fragment (viral backbone) which is deleted of left-end elements required for virus replication and packaging. Generating such a fragment from Ad7a is complicated by the fact that the majority of the Ad7a DNA sequence has not been determined (7,8,15). By analyzing the available Gomen strain Ad7 left-end sequence, we identified a battery of restriction endonucleases which were able to digest Ad7 DNA at an appropriate site at the left end.…”

“…It is deduced from these observations that EBB gene products play an important role in the transformation of cells. DNA sequence data of the EBB gene reveal that there are at least two polypeptides encoded by EBB (19,000-molecular-weight [19K] and 54K polypeptides in Adl2) (3,4,16,25,29). The smallest Adl2 DNA fragment capable of transforming rodent cells completely is the HindIII-G (0 to 6.8 map units) (22).…”
