The Nox1/Nox4 inhibitor attenuates acute lung injury induced by ischemia-reperfusion in mice

Abstract: Lung ischemia and reperfusion injury (LIRI) were mediated by several processes including over-production of reactive oxygen species (ROS) and inflammatory activation. ROS generated by nicotinamide adenine dinucletide phosphate (NADPH) oxidase (Nox) may play a pivotal role in pathophysiological changes in a range of disease. However, it was poorly understood in LIRI. Thus, the purpose of our study was to explore whether GKT137831, as a special dual inhibitor of Nox1 and 4, could alleviate LIRI in mice model and… Show more

“…However, the role of NOX in autophagy regulation appears to be dependent on the disease pathology. For example, increases in NOX1 (and NOX4) have been shown to be involved in increase in autophagy associated with HI injury [ 93 ], in transport stress in the pig jejunum [ 94 ], and in the decrease in angiogenesis associated with persistent pulmonary hypertension of the newborn (PPHN) [ 95 ]. Further, in the latter study a positive feedback relationship between autophagy and NOX activity was identified [ 95 ].…”

“…GKT137831, an inhibitor of NOX1 and NOX4, is under clinical trial and has been reported to reduce oxidative stress and diabetic vasculopathy [ 272 ]. GKT137831 has been reported to protect lung tissue damage after ischemia and reperfusion injury in a murine model [ 273 ]. Nox2 inhibitors (CPP11G and CPP11H) has proven to counteract ROS production by inhibiting stress-responsive MAPK signaling and downstream AP-1 and NF-κB nuclear translocation in human ECs.…”

“…In addition, Nox4 interacts with toll like receptor (TLR)-4 inducing NF-κB activation and inflammation (70). Taken together, these effects may contribute to increased ARDS severity, and inhibition of Nox-4 attenuated ALI in mice (71). Oxidative damage plays a significant role in agerelated disease severity.…”
