The Non-Peptide Arginine-Vasopressin v1a Selective Receptor Antagonist, SR49059, Blocks the Rewarding, Prosocial, and Anxiolytic Effects of 3,4-Methylenedioxymethamphetamine and Its Derivatives in Zebra Fish

Abstract: 3,4-Methylenedioxymethamphetamine (MDMA) and its derivatives, 2,5-dimethoxy-4-bromo-amphetamine hydrobromide (DOB) and para-methoxyamphetamine (PMA), are recreational drugs whose pharmacological effects have recently been attributed to serotonin 5HT2A/C receptors. However, there is growing evidence that the oxytocin (OT)/vasopressin system can modulate some the effects of MDMA. In this study, MDMA (2.5–10 mg/kg), DOB (0.5 mg/kg), or PMA (0.005, 0.1, or 0.25 mg/kg) were administered intramuscularly to adult zeb… Show more

“…Four studies examined ASD-specific behaviour in MDMA-treated fish, in particular social behaviour [ 103 - 106 ]. They found that when 1 or 5 mg/kg MDMA was singularly intramuscularly injected into electric fish, asocial behaviour decreased and prosocial behaviour increased [ 103 ].…”

“…This is because fish tend to become anxious towards the water’s surface as there is a greater risk of predation there [ 108 , 109 ], and shoaling behaviour is generally a result of alarm pheromones dissipated among the fish [ 104 , 110 ]. Ponzoni et al (2016, 2017) found that a singular intramuscular injection of 0.1-10 mg/kg MDMA increased social preference [ 105 , 106 ]. Two studies looked at social behaviour in singularly MDMA-immersed octopodes [ 111 , 112 ].…”

“…The above-mentioned AVPV 1A Rs and AVPV 1B Rs distinctions suggest that a drug-induced reward in rodents may be attenuated by the blockage of AVPV 1B Rs, whereas the blockage of AVPV 1A Rs prevents the amelioration of a drug reward. Interestingly, the SR49059-induced AVPV1ARs blockage prevented the acquisition of MDMA-CPP in adult zebrafish [ 98 ], indicating a possible species-dependent effect; however, available data are insufficient to fully confirm this hypothesis. The effects of AVP and AVPRs ligands on behavioral/rewarding effects of different drugs of abuse has been summarized in Table 3 and Table 4 , respectively.…”
