The NLRP3 Inflammasome Pathway: A Review of Mechanisms and Inhibitors for the Treatment of Inflammatory Diseases

Abstract: The NLRP3 inflammasome is a multiprotein complex that plays a pivotal role in regulating the innate immune system and inflammatory signaling. Upon activation by PAMPs and DAMPs, NLRP3 oligomerizes and activates caspase-1 which initiates the processing and release of pro-inflammatory cytokines IL-1β and IL-18. NLRP3 is the most extensively studied inflammasome to date due to its array of activators and aberrant activation in several inflammatory diseases. Studies using small molecules and biologics targeting th… Show more

“…Dysfunctional RPE cells by oxidative stress drives inflammation. ROS in RPE cells promote NF-κB signaling and nod-like receptor family pyrin domain containing 3 (NLRP3) inflammasome [ 36 , 37 ], which further upregulates IL-1β and IL-18 through the activation of caspase-1 [ 38 ]. ROS is also associated with angiogenesis in the retina and choroid [ 39 ].…”

“…This requires a system that is tightly regulated to allow for activation but also to ensure a limited amount of collateral damage. Dysregulation of the inflammasome can result in aberrant or excessive stimulation, contributing to pathologies related to auto-inflammatory conditions, cancer, cardiometabolic disorders, and neurodegenerative diseases (Anderson, Biggs, Rankin, & Havrda, 2022;Blevins, Xu, Biby, & Zhang, 2022;Boxberger, Hecker, & Zettl, 2019;Holbrook et al, 2021). Thus, genetic or environmental conditions that stimulate, alter, or disrupt inflammasome activation can have significant effects upon the organism.…”

“…The NACHT, LRR, and PYD domain-containing protein 3 (NLRP3) inflammasome is a multiple protein complex, consisting of NLRP3 protein as a sensor, apoptosis-associated speck-like protein (ASC) as an adaptor and caspase-1 as an effector. 81 Typically, the assembly of NLRP3 inflammasome requires a priming stage and activation. The priming of the formation of NLRP3 inflammasome needs upstream signals as the first signal from the engagement of pathogenassociated molecular patterns (PAMPs) or damage-associated molecular patterns (DAMPs) with TLR, NOD2, IL-1R, or tumor necrosis factor receptor (TNFR), which subsequently triggers the upregulation of NLRP3 and pro-inflammatory cytokines: pro-interleukin (IL)-1β and pro-IL-18 via NF-κB pathway.…”
