The Nine Genes of the Nocardia lactamdurans Cephamycin Cluster Are Transcribed into Large mRNAs from Three Promoters, Two of Them Located in a Bidirectional Promoter Region

Abstract: The nine biosynthesis genes of the Nocardia lactamdurans cephamycin cluster are expressed as three different mRNAs initiating at promoters latp, cefDp, andpcbABp, as shown by low-resolution S1 nuclease protection assays and Northern blotting analysis. Bidirectional expression occurred from divergent promoters (latp andcefDp) located in a 629-bp intergenic region that contains three heptameric direct repeats similar to those recognized by members of the SARP (Streptomyces antibiotic regulatory proteins) family.… Show more

“…Coupling to the xylE reporter gene. To study if expression from latp and pcbABp was regulated by diaminopropane, DNA fragments containing each promoter (11) were coupled in the appropriate orientation to the xylE in the promoter-probe vector pIJ4083 (3).…”

“…The N. lactamdurans genes encoding cephamycin biosynthesis enzymes have been cloned and sequenced and shown to be clustered (6) in a 30-kb region of the genome of this microorganism (22). The genes of the cephamycin cluster are expressed as polycistronic mRNAs from three promoters, latp, pcbABp, and cefDp (11).…”

“…Genes belonging to the same metabolic pathway are very often found clustered and organized in operons in bacterial genomes. Examples of gene clusters resulting in large polycistronic transcripts 17.5 and 16 kb have been previously described in Clostridium difficile (Hammond et al, 1997) and Nocardia lactamdurans (Enguita et al, 1998) respectively. This is also true for the genes involved in photosynthesis (PS).…”

“…The ribosome slow-down may detach the RNA polymerase from the DNA reducing expression of the ceaS2 gene and modulating expression of the ccaR and claR genes. Attenuation of antibiotic resistance genes as a mechanism of control of gene expression was proposed a few years ago by Weisblum and coworkers (Mayford and Weisblum, 1990;Kwak et al, 1991) but this mechanism has not been studied in detail, although mechanisms of attenuation (termination/ antitermination) were proposed to explain the different overlapping transcripts of the cephamycin gene cluster (Enguita et al, 1998). The rplK gene is linked to nusG encoding a transcriptional antiterminator-like protein in different Streptomyces species and is, at least in part, cotranscribed with nusG (J.P. Gomez-Escribano, unpubl.…”
