The new ophthalmic formulation for infection control by combining collagen/gelatin/alginate biomaterial with liposomal chloramphenicol

Abstract: Eye drops are a conventional method of drug delivery to the eye, accounting for 90% of currently accessible ophthalmic formulations. The major problem with eye drop treatments is rapid pre-corneal drug loss. Furthermore, the need for frequent administration of eye drops can profoundly affect the quality of life of ophthalmological patients. In the current study, we developed a liposomal nanoparticle encapsulated with chloramphenicol mixed with biodegradable materials against ophthalmological disease. We first … Show more

“…Alginate is an anionic copolymer with Lguluronic acid and D-mannuronic blocks acid [93], which has been also extensively used to construct biocompatible hydrogels. Some studies demonstrated that alginate might induce myocardial repair and regeneration, and stem cell delivery [94,95]. Alginate could quickly react to divalent cations (e.g.…”

“…Our first step at this stage was to establish the appropriate formulation of CGA-Lipo-MFX/DEX for ophthalmic use. Based on our degradation curve measurement, we developed a formula to determine the relationship between the molar ratio of sodium citrate to sodium alginate with the CGA decomposition time: Y = 12.3314X 2 − 38.6691X + 29.2180 (where Y is the CGA decomposition time, and X is the molar ratio of sodium citrate to sodium alginate) [24]. Based on this degradation formula, we designed an 8 h degradable formulation by adjusting the sodium citrate/sodium alginate ratio in our investigation.…”

“…Based on our previous investigation, we established the formula to determine the relationship between the molar ratio of sodium citrate to sodium alginate with CGA decomposition time [24]. The CGA complex completely degraded in 0.01% sodium citrate.…”

“…Therefore, apart from designing the vehicle/base composition, drugs are often incorporated into appropriate carriers or introduced into systems whose purpose is to provide the expected concentration in the treated tissue for the desired time period. The most frequently studied and described are liposomes [60,61], micelles [60,62], microparticles [63][64][65], nanoparticles [66,67], micro- [68,69], and nanoemulsions [70,71]. Unfortunately, it has to be noticed that most technological solu-tions on the nanoscale are only promising at the laboratory stage.…”
