The myb-related gene stonewall induces both hyperplasia and cell death in Drosophila: rescue of fly lethality by coexpression of apoptosis inducers

Abstract: We carried out gain-of-function mutagenesis screening and identified a mutant in which GAL4 induction led to both hyperplasia and apoptosis. The gene involved was identified as stonewall (stwl), a myb-related gene involved in germ cell proliferation and differentiation during oogenesis. As observed with dmyb, the ectopic expression of stwl UY823 inhibited endoreplication in salivary glands. We also found that stwl UY823 overexpression, like overexpression of the wildtype gene, activated G1/S transition and apo… Show more

“…Moreover, we found several proteins involved in the apoptotic pathway, as the transcription factor Stwl 23 , the mitochondrial carrier homolog 1 (Mtch) 24 , ALG-2 interacting protein X 21 and the GTPase Rab1 25 , 26 . Among these proteins, we focused our attention on the ALG-2 interacting protein X, also called Alix.…”

“…All three of these pathways converge on Bim, a pro-apoptotic protein that is activated in response to NGF deprivation. Overexpression of either FOXO or Myb family proteins (including STWL) can result in apoptotic cell death (Liu and Greene 2001;Gilley et al 2003;Brun et al 2006). However, the apoptotic response to NGF under physiological conditions may require the combined derepression of all three of these pathways (Biswas et al 2007).…”
