The multiplex bead array approach to identifying serum biomarkers associated with breast cancer

Abstract: Introduction Breast cancer is the most common type of cancer seen in women in western countries. Thus, diagnostic modalities sensitive to early-stage breast cancer are needed. Antibodybased array platforms of a data-driven type, which are expected to facilitate more rapid and sensitive detection of novel biomarkers, have emerged as a direct, rapid means for profiling cancer-specific signatures using small samples. In line with this concept, our group constructed an antibody bead array panel for 35 analytes tha… Show more

“…This adhesion protein has also been identified as a serum marker in other types of tumors including breast cancer37 and hepatocellular carcinoma 38. VN is a glycoprotein involved in cell adhesion, extracellular matrix remodeling and in tumor cell migration, principally through its interaction with integrins, the urokinase‐type plasminogen activator receptor (uPAR) and plasminogen activator inhibitor‐1 (PAI‐1) 39. VN has not been studied as a serum marker associated with melanoma prognosis to date; however, VN receptor expression on melanoma cells has been associated with poor disease prognosis 15, 40.…”

“…This protein has been noted as having a protective effect in breast cancer by facilitating transport of vitamin D metabolites (13) and has been shown to be downregulated in the serum of breast cancer patients (26). Some studies suggest that specific genetic variants of VDBP influence a woman's risk of developing breast cancer (27).…”

“…More detailed information can be obtained from elsewhere (Angenendt et al, 2006;Diamandis, 2003;Haab, 2005;Kim et al, 2009;Petricoin et al, 2002). A relatively new proteomic technique that is suitable for high-throughput profiling is Surface Enhanced Laser Desorption/Ionization-Time-Of-Flight (SELDI-TOF) which combines affinity chromatography on a modified surface (surface-enhanced) with MALDI-TOF.…”
