The multifaceted role of the SASP in atherosclerosis: from mechanisms to therapeutic opportunities

Abstract: Background The global population of older individuals is growing, and ageing is a key risk factor for atherosclerotic cardiovascular diseases. Abnormal accumulation of senescent cells can cause potentially deleterious effects on the organism with age. As a vital marker of cellular senescence, the senescence-associated secretory phenotype (SASP) is a novel mechanism to link cellular senescence with atherosclerosis. Main body In this review, we concr… Show more

“…Inflammaging is considered a causal risk factor for cardiovascular diseases [ 1 , 2 ]. Throughout life, senescent cells accumulate in vascular tissue, resulting in the development of inflammaging, which consequently leads to cardiovascular diseases [ 1 , 2 , 3 , 4 ]. Senescent cells have been observed in atherosclerotic tissue, and by eliminating senescent cells, the progression of atherosclerosis could be prevented [ 1 , 2 , 5 ].…”

“…Senescent cells have been observed in atherosclerotic tissue, and by eliminating senescent cells, the progression of atherosclerosis could be prevented [ 1 , 2 , 5 ]. The abolition of senescent cells has been shown to increase lifespan and provide protection against cardiovascular and age-related diseases in experimental animal studies [ 3 , 6 ].…”

“…Instead, extracellular vesicles, such as exosomes, may serve as cell-free therapeutic agents. Not only were they not at risk of promoting tumourigenesis, but they were also less susceptible to immune rejection ( 228 ). An intriguing possibility is that exosomes may carry biological information which may be beneficial or detrimental, suggesting they can function as anti-aging agents ( 229 ).…”

“…In particular, the Nod-like receptor protein 3 (NLRP3) inflammasome can be primed or indirectly activated by inflammatory cytokines (e.g. IL-1β, IL-6) that are often found as components of the SASP (38,(54)(55)(56).…”
