The Multi-Faceted Effect of Curcumin in Glioblastoma from Rescuing Cell Clearance to Autophagy-Independent Effects

Abstract: The present review focuses on the multi-faceted effects of curcumin on the neurobiology glioblastoma multiforme (GBM), with a special emphasis on autophagy (ATG)-dependent molecular pathways activated by such a natural polyphenol. This is consistent with the effects of curcumin in a variety of experimental models of neurodegeneration, where the molecular events partially overlap with GBM. In fact, curcumin broadly affects various signaling pathways, which are similarly affected in cell degeneration and cell di… Show more

“…Sci. 2021, 22, 8566 2 of 26 (GBM) [6]. To circumvent its poor oral absorption following the inclusion of curcumin in the diet, other strategies have been proposed to treat experimental models of GBM or malignant mesothelioma (MM), e.g., intraperitoneal injection [6,7].…”

To circumvent its poor oral absorption following the inclusion of curcumin in the diet, other strategies have been proposed to treat experimental models of GBM or malignant mesothelioma (MM), e.g., intraperitoneal injection [6,7]. Based on proteomic analyses of the liver from curcumin-treated tumor-bearing rats vs. untreated tumor-bearing rats and normal rats, we previously identified quantitative changes in a set of biomarkers representing both determinants of liver colonization and therapeutic targets [8].…”

“…De acordo com a revisão realizada por Ryskalin et al (2020) na verdade, a curcumina afeta amplamente várias vias de sinalização, que são afetadas de forma semelhante na degeneração celular e na diferenciação celular. Os efeitos antitumorais da curcumina incluem inibição do crescimento, parada do ciclo celular, anti--migração e anti-invasão, bem como atividade quimio e radio-sensibilizante.…”

“…The BBB permeable nutraceutical curcumin has shown to exert anti-proliferative effects on glioma cells by modulating TP53/MDM2/MDMX/p14ARF signaling. In particular, curcumin upregulates p53 expression in GB in vitro and induces cell cycle arrest in a p53-dependent manner ( 167 , 206 ). Pre-clinically in GB, chalcone derivatives and flavopiridol have shown to decrease MDM2 protein level or inhibit MDM2 expression at mRNA level, respectively ( 168 , 207 ).…”
