The MP65 gene is required for cell wall integrity, adherence to epithelial cells and biofilm formation in Candida albicans

Abstract: BackgroundThe MP65 gene of Candida albicans (orf19.1779) encodes a putative β-glucanase mannoprotein of 65 kDa, which plays a main role in a host-fungus relationship, morphogenesis and pathogenicity. In this study, we performed an extensive analysis of a mp65Δ mutant to assess the role of this protein in cell wall integrity, adherence to epithelial cells and biofilm formation.ResultsThe mp65Δ mutant showed a high sensitivity to a range of cell wall-perturbing and degrading agents, especially Congo red, which i… Show more

“…There is an adhesin-like protein expressed at highest levels in yeast cells, Ywp1 ( Y east W all P rotein 1), but it seems to function as an anti-adhesin (38). There are also proteins that may function as adhesins but lack a GPI anchor, including Mp65 ( M anno p rotein of 65 kDa) (39), Csh1 ( C ell S urface H ydrophobicity) (40) and Pra1 ( p H r egulated a ntigen) (32). …”

“…Of these differential responses in the two biofilm phenotypes, a vast number of genes upregulated in the Agg biofilm belonged to CC as assessed using GO analyses Specifically, several cell wall genes were upregulated in the Agg phenotype e.g., TSA1 , ECM33 , MP65 , ALS1 and PHR1 . In C. albicans , ECM33 and MP65 have been implemented as important proteins in maintenance of fungal cell wall integrity, biofilm formation and stress responses [4143]. Furthermore, the ALS family of adhesin proteins and PHR family of extracellular transglycosylases also function as important regulators of biofilm formation in C. albicans [44, 45].…”

“…Primer3 software was used to design ERG3 primers. Primers for MP65 and ACT1 were obtained from the study of Sandini et al (Table 1) [8]. The actin producing gene, ACT1 was employed in the current investigation as the housekeeping gene.…”

“…Changes in MP65 gene which encodes a cell wall mannoprotein by tunicamycin also might affect cell wall organization and has been suggested as an attractive potential target for the development of antifungal drugs [8]. Previous studies indicate that MP65 gene appears in cell wall mannoprotein synthesis (Mp65p) and stability [8] which is then translated into fungal virulence.…”
