The mouse SWISS-2D PAGE database: a tool for proteomics study of diabetes and obesity

Abstract: A number of two-dimensional electrophoresis (2-DE) reference maps from mouse samples have been established and could be accessed through the internet. An up-to-date list can be found in WORLD-2D PAGE (http://www.expasy.ch/ch2d/2d- index.html), an index of 2-DE databases and services. None of them were established from mouse white and brown adipose tissues, pancreatic islets, liver nuclei and skeletal muscle. This publication describes the mouse SWISS-2D PAGE database. Proteins present in samples of mouse (C57B… Show more

“…In general, massive results have been obtained preferentially for organisms whose genome has been enterely sequenced. This is the case of microrganisms presenting a reduced genome in size or the poor number of multicellular species that have chosen for their importance or as model systems (H. sapiens, M. musculus, P. falciparum, C. elegans, D. melanogaster and O. sativa) [7][8][9][10][11][12][13].…”

“…From the 2D PAGE database, the pI value for HBB and the hemoglobin "-chain was found to be around 7.5 and 9.0, respectively. 25,26 The same experiments were conducted using a HepG2 cell line. As shown in Figure 4, the pattern for the bands of primary parenchymal cells from mice livers stained with CBB was quite different from the pattern obtained for the HepG2 cells, whereas the band was similarly obtained at around 16 kDa.…”

“…Also, in order to confirm spot identity, these techniques should be compatible with MS. It is also desirable that the separation pattern from alternative techniques is analogous to the one obtained in 2-DE to allow for the further use of the comprehensive and power- [44,45]. These techniques may include capillary isoelectric focusing coupled to mass spectrometry (CIEF-MS n ) and preparative isoelectric focusing followed by size-exclusion chromatography, hyphenated with MS (PIEF-SEC-MS), which shall be thoroughly discussed in the main section.…”
