The Morphoregulatory Role of Thidiazuron: Metabolomics-Guided Hypothesis Generation for Mechanisms of Activity

Abstract: Thidiazuron (TDZ) is a diphenylurea synthetic herbicide and plant growth regulator used to defoliate cotton crops and to induce regeneration of recalcitrant species in plant tissue culture. In vitro cultures of African violet thin petiole sections are an ideal model system for studies of TDZ-induced morphogenesis. TDZ induces de novo shoot organogenesis at low concentrations and somatic embryogenesis at higher concentrations of exposure. We used an untargeted metabolomics approach to identify metabolites in co… Show more

“…Despite more than 40 years of use, universal application in the environment and hundreds of scientific studies demonstrating TDZ-induced plant morphogenesis, the precise mechanism of action remains unknown. Recently, using a metabolomics approach, several hypotheses for the mechanism of action of TDZ were suggested for understanding its regulatory role in plant morphogenesis [ 65 ].…”

“…In the synthetic biotransformations or logical algorithms approach developed in our group we use common chemical reactions such as (de) methylation, oxidation, reduction, (de) hydroxylation, (de) glycosylation, (de) carboxylation and apply these to a select subset of metabolites of interest to predict new pathways and metabolites in a sample based on a known starting point. 4,6,11,10 This approach uses the predicted change in monoisotopic mass of a specific metabolite by the addition or removal of a common moiety which can then be mined in the metabolomics dataset.…”

“…5 This approach was then expanded and adapted to an untargeted metabolomics work flow. 4,6 Hormones are not however exclusive to plants and in fact are best studied in mammalian systems having been defined as "substance [s] which, being produced in any one part of the organism, is transferred to another part and there influence a specific physiological process". 7 Hormones are often hard to quantify due to their labile nature and presence at low concentrations.…”
