The Monoclonal Antibody against the Major Diagnostic Antigen ofParacoccidioides brasiliensisMediates Immune Protection in Infected BALB/c Mice Challenged Intratracheally with the Fungus

Abstract: The protective role of specific antibodies against Paracoccidioides brasiliensis is controversial. In the present study, we analyzed the effects of monoclonal antibodies on the major diagnostic antigen (gp43) using in vitro and in vivo P. brasiliensis infection models. The passive administration of some monoclonal antibodies (MAbs) before and after intratracheal or intravenous infections led to a reduced fungal burden and decreased pulmonary inflammation

“…The count of fungal cell viability, expressed as colony forming units (CFU), was used to determine the survival rate of melanized and nonmelanized yeast when exposed to oxidants. Intratracheal infection of BALB/c mice with 3x10 5 yeast cells of P. brasiliensis, melanized and nonmelanized, was performed as previously described (Buissa-filho et al 2008). For statistical analysis, the GraphPad Prism5 software was utilized.…”

“…In patients that underwent chemo-therapy, both gp43 and gp70 are markers for monitoring successive treatment and cure through their decreased antigenemia and specific antibody response (Marques da Silva et al 2004. In the experimental Balb/c model of PCM infection, anti-gp43 mAb 3E effectively reduced the fungal burden and promoted phagocytosis in vitro (Buissa-Filho et al 2008). The recognized epitope in the gp43 was mapped to the sequence NHVRIPIGYWAV shared with Aspergillus fumigatus, A. oryzae and B. graminis internal sequences of β-1,3-glucanases.…”

“…IgG2a and IgG2b MAbs to gp43 of P. brasiliensis were shown to reduce fungal burden and was associated with the enhanced phagocytosis of P. brasiliensis by macrophages leading to increased nitric oxide production [82]. Prophylaxis passive intranasal administration of anti-glycoprotein A IgM or IgG1 switch variant MAb protected against murine P. carinii [83].…”
