Abstract: The short‐chain dehydrogenase/reductase (SDR) superfamily members acyl‐ACP reductases FabG and FabI are indispensable core enzymatic modules and catalytic orientation controllers in type‐II fatty acid biosynthesis. Herein, we report their distinct substrate allosteric recognition and enantioselective reduction mechanisms.
