The Mitochondrial Ornithine Transporter

Abstract: Two isoforms of the human ornithine carrier, ORC1 and ORC2, have been identified by overexpression of the proteins in bacteria and by study of the transport properties of the purified proteins reconstituted into liposomes. Both transport L-isomers of ornithine, lysine, arginine, and citrulline by exchange and by unidirectional mechanisms, and they are inactivated by the same inhibitors. “…For the closely related sequence of Ndt2p, which was also cloned and expressed in E. coli in this study, no biochemical data are available, because we were incapable of renaturing and/or reconstituting it functionally. However, because of the high degree of homology between Ndt1p and Ndt2p (70% homology and 55% identical amino acids), which is similar to that found for other mitochondrial carrier isoforms (33,39,45), it is likely that Ndt2p is an isoform of the NAD ϩ transporter in S. cerevisiae.
