The miR-126–VEGFR2 axis controls the innate response to pathogen-associated nucleic acids

Abstract: MicroRNA-126 (miR-126) is a microRNA predominately expressed by endothelial cells and controls angiogenesis. We found miR-126 was required for the innate response to pathogen-associated nucleic acids, and that miR-126-deficient mice had increased susceptibility to pseudotyped-HIV infection. miRNA profiling and deep-sequencing indicated that miR-126 was highly and specifically expressed by plasmacytoid dendritic cells (pDCs). miR-126 controlled the survival and function of pDCs, and regulated expression ofinnat… Show more

“…Recently, Marti et al showed that VEGF can not only regulate DC maturation but also induce immunosuppressive phenotype through the production of IDO [61]. Although plasmacytoid DC (pDC) function in cancer remains unclear, Agudo et al recently reported the role of VEGF signaling in pDC maturation in lymphoid tissues, which is mediated through an miRNA, miR-126 [62]. They showed that miR-126 is controlling the number of pDCs in secondary lymphoid organs and is regulating the immune function of pDCs, including type I interferon production.…”

“…miR150 is required for the crosspresentation capacity of Langerhans cells (skinresident DCs) [45] . In addition, miR29b, miR 29c [46] , miR126 [47] , miR146a [40,48] , miR155, and miR221 [49] have been shown to regulate DC apoptosis and cytokine production.…”

“…Bars represent mean values and standard deviations based on triplicate experiments (**, P Ͻ 0.01). (45). There is a body of vast literature on miR-155 involvement in immunity and inflammation, with one proposed mode of action being a positive correlation between miR-155 upregulation and NF-B activation (46).…”
