The migration speed of cancer cells influenced by macrophages and myofibroblasts co-cultured in a microfluidic chip

Abstract: We employ a microfluidic chip with three culture chambers to investigate the interactions among lung cancer cells, macrophages and myofibroblasts. By mixing the conditioned media of macrophages and myofibroblasts in this chip, we confirm that these two stromal cells have synergistic effects in accelerating the migration of cancer cells. However, as the myofibroblasts are pretreated with the conditioned medium of macrophages, the myofibroblasts' ability to enhance the migration of cancer cells is lowered. The t… Show more

“…Interestingly, instead of pretreatment, direct combination of macrophage conditioned medium and myofibroblast conditioned medium resulted in a nearly threefold increase of lung cancer cells' migration speed [167]. Similar to other triculture microfluidic models, these results imply that the responses of cancer cells influenced under multiple factors can be quite complex and diverse [155,167].…”

“…In our opinion, the overlook of tumor microenvironment has inescapable relationship with tough problem. In vivo, cancer cells were cultured in dishes in conventional methods such as transwell assay 16 or bathing cells in conditioned medium 17 , but with poor in vivo-like microenvironment reconstruction 18,19 . In the present study, a 3D microfluidic device was developed to reconstruct an in vivo-like tumor microenvironment for bladder cancer cells.…”

“…Although a few methods such as transwell assay 16 or bathing cells in conditioned medium 17 were proposed to study the interactions between cancer cells and stromal cells, most of them are still limited because of the poor in vivo-like microenvironment reconstruction 18,19 . Recently, microfluidics has attracted intensive attention in molecular and cellular biology research due to its miniaturized size, high throughput detection, low consumption of reagents and especially the accurate reconstruction of 3D in vivo-like microenvironment 1,13 .…”
