Alpha (α)-tubulin (TUA) is a primary unit of cytoskeleton microtubules. In recent years, more frequent frosts have caused considerable yield loss in sugarcane (Saccharum spp. hybrid) production. Temperature is one of the most influential abiotic factors affecting the microtubule polymerization state in plants. In this research, we developed overexpressing SoTUA transgenic sugarcane lines from the cold-susceptible cultivar ROC22. The transgene was confirmed by polymerase chain reaction (PCR) and quantificational real-time polymerase chain reaction (qRT-PCR) analysis, while the relative expression of SoTUA protein was verified by Western blot. Compared with the nontransformed control plants (wild type [WT]), the transgenic lines showed higher relative protein expression than the WT at 25 ˚C, which indicates the overexpression of SoTUA protein. The contents of soluble protein and sugar and the activity of peroxidase (POD) were higher while the content of maleicdialdehyde was lower in the transgenic plants than the WT plants under the chilling treatment. Meanwhile, the expression levels of cold-defense related genes (P5CS, WRKY, Cu/Zn-SOD, and POD1) were higher in the transgenic lines than in the WT. These results suggest that the SoTUA protein plays an important role in protecting plants during chilling stress. This positive effect is not affected by transgenic plant generations.

The detyrosination/retyrosination cycle is the most common post-translational modification of α-tubulin. Removal of the conserved C-terminal tyrosine of αtubulin by a still elusive tubulin tyrosine carboxypeptidase, and religation of this tyrosine by a tubulin tyrosine ligase (TTL), are probably common to all eukaryotes. Interestingly, for plants, the only candidates qualifying as potential TTL homologs are the tubulin tyrosine ligase-like 12 proteins. To get insight into the biological functions of these potential TTL homologs, we cloned the rice TTL-like 12 protein (OsTTLL12) and generated overexpression OsTTLL12-RFP lines in both rice and tobacco BY-2 cells. We found, unexpectedly, that overexpression of this OsTTLL12-RFP increased the relative abundance of detyrosinated α-tubulin in both coleoptile and seminal root, correlated with more stable microtubules. This was independent of the respective orientation of cortical microtubule, and followed by correspondingly changing growth of coleoptiles and seminal roots. A perturbed organization of phragmoplast microtubules and disoriented cell walls were further characteristics of this phenotype. Thus, the elevated tubulin detyrosination in consequence of OsTTLL12 overexpression affects structural and dynamic features of microtubules, followed by changes in the axiality of cell plate deposition and, consequently, plant growth.

Influence of exogenous abscisic acid on alterations in protein expression in the proteome of Triticosecale seedlings

The aim of this study was to investigate molecular basis of germination inhibition of Triticosecale under the influence of exogenous ABA. Seedlings were isolated from seeds after 48 h of germination in water (control sample) and 100 lM ABA solution. It was observed that the applied concentration of phytohormone caused a significant inhibition of germination and a suppressed contribution of polysomes in the total ribosomal fraction. To identify proteins whose expression was affected by ABA, a proteomic approach employing 2D electrophoresis was used. Proteome maps for both control and ABA-treated samples displayed over 2,200 Coomassie Brilliant Blue-stained spots each. Protein spots showing either increase or decrease in spot intensity under the ABA influence were identified by mass spectrometry. Many of the identified proteins whose expression was stimulated by ABA proved to be stress-related, involved in nucleotide metabolism or playing a regulatory and signal transduction role, whereas proteins whose expression decreased are known to be involved in numerous metabolic pathways (including energy, carbohydrate or nucleotide metabolism) as well as related to genetic information processing and protein synthesis. Our results show that the germination inhibition caused by ABA is a result of multigene, diversified actions leading together to triticale growth suppression.
