The Methodology Applied in DPPH, ABTS and Folin-Ciocalteau Assays Has a Large Influence on the Determined Antioxidant Potential

Abstract: Antioxidant potential (AOP) is not only the property of the matrix analyzed but also depends greatly on the methodology used. The chromogenic radicals 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (and Folin-Ciocalteu (FC) assay were applied to estimate how the method and the composition of the assay solvent influence the AOP determined for coffee, tea, beer, apple juice and dietary supplements. Large differences between the AOP values depending on the reaction medium were observed, with the highest A… Show more

“…Caffeic acid and its ester (CGA) have similar yet kinetically different profiles. On the contrary to GA, contribution of secondary reactions in DPPH assay at pH 7.4 is We have observed the sample speci c kinetic pro les also for di erent food matrices [22]. e dΔA/dt on the time scale of few tens of minutes when samples as tea, co ee, cranberry, and apple juice were analyzed was even larger than observed for antioxidant compounds in this study.…”

“…AOP of food samples is usually evaluated by more than one method, and correlation analysis of AOPs obtained by DPPH, ABTS, and FC assays is often performed. e correlations can be high and significant or weak [20][21][22], reflecting the lack of consistency of the results of such analyses. e large influence of the experimental parameters on the reactivity of antioxidants in the samples undoubtedly contributes to the observed discrepancies.…”

“…Therefore, it is impossible to determine exactly the type and the amount of the contained antioxidants. The use of various tests can instead lead to results that are able to differentiate the whole matrice in their heterogeneity and complexity and better describe their nutritional values (Shahidi & Ambigaipalan, 2015;Shahidi & Zhong, 2015;Abramovi c et al, 2017).…”

“…The first step to quantify the antioxidant activity of a plant extract is to select the right method [9]. There is a large variety of methods to determine this parameter [10], and the variability of experimental conditions found in the literature for each of the methods hinders such selection and the possibility of easily comparing the obtained results with those of other authors.…”
