The metabolism of benzyl isothiocyanate and its cysteine conjugate in guinea-pigs and rabbits

Abstract: 1. Following oral administration of benzyl isothiocyanate or its cysteine conjugate the major metabolite isolated from urine of guinea-pigs and rabbits was a cyclic mercaptopyruvate conjugate, 4-hydroxy-4-carboxy-3-benzylthiazolidin-2-thione. 2. Renal excretion of the metabolite after oral administration of benzyl isothiocyanate or its cysteine conjugate to guinea-pigs amounted to 23% and 33% respectively. The corresponding N-acetylcysteine conjugate of benzyl isothiocyanate (a mercapturic acid) was present as… Show more

“…The cyclic mercaptopyruvic acid (4-hydroxy-4-carboxy-3-benzylthiazolidin-2-thione) occurred in the urine from guinea-pigs and rabbits after the oral or intravenous administration of benzyl isothiocyanate or its cysteine metabolite. 39 In human individuals, the isothiocyanates are mainly metabolized via the mercapturic acid pathway. 40 However, to the best of our knowledge, mercaptopyruvic acid has not been observed in human intervention studies up to now, and we also did not detect it in our study.…”

“…These studies show that guinea pigs and rabbits do not excrete mercapturic acids during the metabolism of ITCs. The authors pointed out that these species also do not readily form mercapturic acids after administration of other types of xenobiotics [128].…”

“…While cysteine conjugates of ITCs can be absorbed intact [177,191], GSH conjugates were shown to require release of free ITC prior to absorption [187]. Diindolylmethane (DIM) and 2,3-BII, the main hydrolysis products of indole-GLS derived products were detected by De Kruif et al [160] in the liver of rats, suggesting that they are absorbed by the small intestine.…”
