Sunmmary1. A method for measuring the rate of production of "IC-labelled adenine nucleotides, including cyclic adenosine 3',5'monophosphate (cyclic AMP), from ["IC]-adenosinetriphosphate (ATP) was developed and used to study the effects of ATP, adenosine diphosphate (ADP) and adenosine 5'-monophosphate (AMP) on the rate of accumulation of cyclic AMP in cell-free preparations of adenyl cyclase from rat brain. 2. The mechanism by which NaF increases cyclic AMP accumulation was studied by comparing its effect on adenine nucleotide metabolism with that of an ATP regenerating system.3. ADP and ATP are potent inhibitors of phosphodiesterase (PDE) and it is the sum of the concentrations of these two nucleotides which controlled the rate of destruction of cyclic AMP. The effect of these nucleotides was significant even in the presence of 6-7 mm theophylline; theophylline itself inhibited PDE only 50-60%. 4. Fluoride ion had no direct effect on PDE but it inhibited the rate of hydrolysis of ADP and ATP and thus indirectly inhibited PDE. The effect of fluoride ion on cyclic AMP accumulation can be explained, at least in part, by this indirect inhibition of PDE.5. Studies on a more purified preparation of adenyl cyclase clearly demonstrated a direct action of NaF on adenyl cyclase.

Control of Epididymal Sperm Motility; An Approach to Male Fertility Regulation

Thermal immobilization was designed as a simple approach to study factors controlling sperm motility. Heating dissected epididymis or isolated epididymal sperm at 48°C to 52°C for 5 min abruptly and completely immobilized the sperm. The thermally immobilized sperm showed some alteration of membrane lipids, as indicated by the response of the surface ATPase to temperature change. However, no qualitative change in the surface negative charge was detectable by the binding of the cells to DEAE‐Sephadex beads. The immobilized cells can metabolize ATP into ADP and AMP similar to the control epididymal sperm. The ATP‐dependent sliding or disintegration of sperm flagella was completely prevented in the immobolized cells. The findings suggested that heat‐treatment disrupted the integrity of the sperm membrane and the motile apparatus, resulting in the loss of sperm motility. The development of post‐testicular antifertility agents should aim at interfering specifically with these components of the sperm.

The effect of hypothyroidism on the noradrenaline-induced accumulation of 3H-cyclic-AMP was measured in rat epididymal adipose tissue. Adipocytes from hypothyroid animals showed a decreased response to noradrenaline 0.02\m=-\0.2 \m=x\10\m=-\5 m, while higher concentrations of the catecholamine seemed to elicit the same response in cells obtained from euthyroid or hypothyroid animals. The diminished sensitivity to noradrenaline was unaltered after the addition of phentolamine. It is suggested that thyroid hormones modulate the catecholamine-induced lipolytic response at least partly through the adenyl cyclase cyclic-AMPN system. Hypothyroidism has been associated with decreased lipolytic response to cate¬ cholamines in rats (Goodman 8c Bray 1966) as well as in man (Rosenqvist 1972). The present study was undertaken in order to investigate a possible defect in the adenyl cyclase cyclic-AMP system in adipocytes from hypothyroid rats. MATERIAL AND METHODSMale Sprague-Dawley rats (90 g) were made hypothyroid by injecting 600 /¿C 1311 intraperitoneally as described (Hyant Sc Whitney 1967). The control group was selected from the same batch of animals but injected with saline. The animals were kept at 22°C. They were fed ad libitum for 4 to 5 weeks until sacrifice by decapitation. The distal portions of the epididymal fat pads were removed. Adipocytes were prepared with collagenase (Rodbell 1964) with minor modifications of the original procedure.
