The Medial Septum Is Insulin Resistant in the AD Presymptomatic Phase: Rescue by Nerve Growth Factor-Driven IRS1 Activation

Abstract: Basal forebrain cholinergic neurons (BFCN) are key modulators of learning and memory and are high energy-demanding neurons. Impaired neuronal metabolism and reduced insulin signaling, known as insulin resistance, has been reported in the early phase of Alzheimer's disease (AD), which has been suggested to be "Type 3 Diabetes." We hypothesized that BFCN may develop insulin resistance and their consequent failure represents one of the earliest event in AD. We found that a condition reminiscent of insulin resista… Show more

“…20 A study of African-Americans found that IRS1 mutations and endocrine disorders caused by obesity synergistically reduce insulin sensitivity, suggesting that IRS1 variability and obesity together become an important predictor of insulin resistance. 21 In this study, we found that IRS1 produced a non-frameshift insertion mutation in the exonic region of O-T2DM patients and that IRS1 mRNA expression levels were up-regulated in O-T2DM patients compared with healthy controls (Log2fold-change = 0.11).…”

“…These results were consistent with our nding that NGF was positively related to glucose levels and insulin secretion in the second trimester. Moreover, NGF could reduce insulin resistance both in vitro and in vivo by activating insulin receptor substrate 1 (IRS1) [22,23]. These data suggest that NGF acts as a protective factor in glucose metabolism and insulin sensitivity and plays a signi cant role in glucose-induced insulin secretion.…”

“…Peptides were purified by means of reverse-phase high-performance liquid chromatography by using a PrepStar 200 model SD-1 chromatography system (Varian, Palo Alto, CA, USA). NGF protein was purified from rodent submaxillary glands, as already reported [38][39][40].…”

“…Cholinergic neurons were harvested from the septal region of E17 Wistar rat embryos, as previously described [39,40] and dissociated cells were seeded as follows: 1.5 × 10 6 cells on poly-l-lysine (Sigma) coated 35 mm plates (BD Falcon, Durham, NC, USA; 353001) for western blotting analyses and 5 × 10 4 cells on glass coverslips in 24-wells plates (BD Falcon; 351147) for immunofluorescence analysis. Cells were grown in Neurobasal (NB) medium supplemented with 2% B27 (Invitrogen) in a 5% CO 2 incubator at 37 • C (ThermoFisher Scientific, Waltham, MA, USA) for 10 days and then used for treatments (DIV10).…”
