The Mechanisms of Ferroptosis and the Applications in Tumor Treatment: Enemies or Friends?

Abstract: Ferroptosis, as a newly discovered non-apoptotic cell death mode, is beginning to be explored in different cancer. The particularity of ferroptosis lies in the accumulation of iron dependence and lipid peroxides, and it is different from the classical cell death modes such as apoptosis and necrosis in terms of action mode, biochemical characteristics, and genetics. The mechanism of ferroptosis can be divided into many different pathways, so it is particularly important to identify the key sites of ferroptosis … Show more

“…Theoretically, FasL- or TRAIL-induced necroptotic death could occur in cells with inactivated cFLIP L when apoptotic pathways are suppressed by FSH signaling, but this hypothetical event has to be confirmed experimentally in vitro and in vivo. Ferroptosis is inhibited by PI3K/AKT signaling [ 215 ] and can be promoted by autophagic degradation of ferritin [ 216 ]. Therefore, it could be prone to occur in combination with autophagy when pre-antral follicles receive insufficient FSH.…”

“…After ICH, ferrous ions overload, GPX4 deficiency, and PLOOHs cannot be cleared in time pointing to the susceptibility of the fenton chemistry. Overload ROS that exceeds the antioxidant capacity of cells leads to an enhanced oxidative stress response, which directly or indirectly damages proteins, nucleic acids, lipids, and other macromolecular substances ( Tan et al, 2022 ). Finally, the membrane is damaged and the cell collapses and dies due to the lipid peroxidation inside the phospholipid of the cell membrane.…”

“…Ferroptosis is a form of iron‐dependent, non‐apoptotic cell death characterized by distinct morphological, biological, and genetic features when compared to other regulated cell death mechanisms like apoptosis, necrosis, and autophagy. 8 Ferroptosis is marked by an excess production of iron‐dependent intracellular oxygen free radicals, resulting in lipid peroxidation and accumulation, ultimately leading to cell membrane rupture. 9 This process is intricately linked to the maintenance of internal environmental homeostasis and the development of various diseases, particularly cancer.…”
