The mechanism of the increase in mitochondrial proton permeability induced by thyroid hormones

Abstract: Three possible mechanisms by which different levels of thyroid hormones in rats might cause the observed sevenfold change in the apparent proton permeability of the inner membrane of isolated liver mitochondria were investigated. (a) Cytochrome c oxidase was isolated from the livers of hypothyroid, euthyroid and hyperthyroid rats and incorporated into liposomes made with soya phospholipids. There was no difference between the proton current/voltage curves of the three types of vesicles. The hormonal effects, t… Show more

“…the activity of the electron transport chain (Brand et al, 1992;Hulbert et al, 1976;Schlame and Hostetler, 1997). Increases in TH levels in response to cold acclimation may drive rapid increases in the activity of aerobic metabolic enzymes, as well as increases that occur over a longer time scale, requiring changes in gene expression.…”

“…Second, thyroxine affects mitochondrial membrane composition (Hulbert, 2000). Thyroxine may affect the amount of inner mitochondrial membrane (Brand et al, 1992), thus resulting in more mitochondrial stain binding. In this scenario, the increase in fluorescence intensity would be due to an increase in inner mitochondria membrane surface area, rather than in mitochondrial content.…”

“…For example, hypothyroid rats have liver mitochondrial membranes containing more unsaturated phospholipid fatty acyl components (Pehowich, 1999) and are inherently less permeable to protons (Brand et al, 1992;Brand et al, 2003). Another goal of this study, therefore, was to examine the effects of dietary PUFA on mitochondrial proton leak in hibernation and to determine how this might relate to the whole-animal effects of PUFA on hibernation performance.…”
