The Measurement of Corticosteroids in Adrenal Vein Blood of Sheep

Abstract: Summary. A double isotope dilution derivative method for determination of aldost( rorif. cortisol and forticosteront' in adrenal vein blood of sheep is described.' ^C-liiiioIIed steroids are iist-d as the indicator and -Ul-act'tit-anhydride as the reagent for the formation of the labelled derivative.Experimental data accumulated from six years' use of the method are diseti-ssetl with .special eniphasi.s on specifii-ity. sensitivity, accuracy and precision.Mean scretion rates of akiosterone, cortisol and cortic… Show more

“…Cortisone is not measured in this system, since it does not compete effectively with [l,2-3H2]corticosterone for binding sites on dog CBG (Murphy, 1967). Since the secretion rate of corticosterone is only about onetenth that of cortisol in the sheep (Bush & Ferguson, 1953;Coghlan, Wintour & Scoggins, 1966), its concentration in peripheral plasma is substantially lower than that of cortisol, and corticosterone thus contributes little to the values determined with CBG.…”

“…Total plasma corticosteroid concentration was measured in each sample by the competitive binding technique of Bassett & Hinks (1969), and since cortisol is the predominant corticosteroid in the plasma of undisturbed sheep (Bush & Ferguson, 1953;Coghlan, Wintour & Scoggins, 1966;Bassett & Hinks, 1969), the results are expressed as cortisol concentration.…”

“…Cortisol secretion rate (FSR) was calculated in the same way as ASR. The sensitivities of both of these assays compares favourably with the double isotope dilution derivative assay, which has been validated for use with adult sheep plasma (Coghlan et al 1966).…”
