The Mammalian Calcium-binding Protein, Nucleobindin (CALNUC), Is a Golgi Resident Protein

Abstract: We have identified CALNUC, an EF-hand, Ca2+-binding protein, as a Golgi resident protein. CALNUC corresponds to a previously identified EF-hand/calcium-binding protein known as nucleobindin. CALNUC interacts with Gαi3 subunits in the yeast two-hybrid system and in GST-CALNUC pull-down assays. Analysis of deletion mutants demonstrated that the EF-hand and intervening acidic regions are the site of CALNUC's interaction with Gαi3. CALNUC is found in both cytosolic and membrane fractions. The membrane pool is tigh… Show more

“…Class 1A Man9-mannosidase is an enzyme involved in the maturation of glycoproteins (63), and CBP1/Hsp47 is a procollagen-binding chaperone involved in procollagen folding and transport (64). Finally, CALNUC (nucleobindin1), a cis-Golgi protein implicated in calcium homeostasis, and its homolog NEFA (nucleobindin2), were present in the fraction (65). The overall composition of the ERGIC fraction confirms its high purity, because it is devoid of marker membrane proteins for other organelles.…”

“…There is a growing consensus that also the Golgi apparatus can function as an agonist-releasable Ca 2ϩ store (6), the importance of which may, however, greatly differ among the different cell types and ranges from relatively unimportant in Drosophila melanogaster S2 cells (7) to very important in renal LLC-PK 1 cells (8). The storage capacity for Ca 2ϩ in the Golgi is greatly increased by specific proteins like Calnuc/nucleobindin (9,10) and members of the CREC family of Ca 2ϩ -binding proteins (reviewed in Ref. 11).…”

“…3) are consistent with the notion that intra-Golgi transport depends upon Ca 2ϩ efflux from inner membrane stores. Several studies have indicated that the Golgi complex may function as a Ca 2ϩ storage organelle (8,(27)(28)(29). Thus it is feasible that Ca 2ϩ from the lumen of the Golgi directly participated in regulating intra-Golgi transport.…”
